Natural Transformation of Plasmid DNA in Escherichia coli in Sterilized Soil Column

چکیده
Horizontal gene transfer allows rapid spread of antibiotic resistance genes1, which in bacteria inhabiting natural aquatic systems occurs by conjugation, transduction, and transformation2. Natural genetic transformation is characterized by the uptake of free DNA by a recipient bacterium, its chromosomal integration or extrachromosomal stabilization, and its expression, which leads to a new phenotype2. Natural transformation is distinct from induced transformation where cells are made competent (the physiologic state, which allows cells to take up DNA by physical, chemical, or enzymatic manipulation.

Background & Objective: Transformation of plasmid DNA into bacterial competent cells is a key technique for molecular cloning. Transformation can be achieved using either chemical or physical methods, e.g., electroporation. The rate of success in these methods depends on experience and attention to method’s details. Therefore, the higher the efficiency and quality of a transformation method, ...
متن کاملTransformation of Escherichia coli mediated by natural phospholipids.
Transformation system for Escherichia coli based upon introduction of plasmid DNA by natural phospholipids has been developed. Transformants are easily obtained by treatment with natural phospholipids such as phosphatidylethanolamine, phosphatidylcoline, and phosphatidylserine, where the presence of MgCl2 or CaCl2 is essential. This method of transformation is applicable not only for small plas...
متن کاملNatural transformation in Campylobacter species.
Growing cells of Campylobacter coli and C. jejuni were naturally transformed by naked DNA without the requirement for any special treatment. Transformation frequencies for homologous chromosomal DNA were approximately 10(-3) transformants per recipient cell in C. coli and 10(-4) in C. jejuni. Maximum competence was found in the early log phase of growth. Campylobacters preferentially took up th...

Background: Crohn's disease and Ulcerative colitis are known as inflammatory bowel disease with high morbidity which are as a result of increasing immune responses to intestinal microbiota in genetically susceptible individuals. The association of adherent invasive Escherichia coli with Crohn's disease in human has been discussed for decades. The principal aim of this study was to assess the re...
متن کاملNatural plasmid transformation in Escherichia coli.
Although Escherichia coli does not have a natural transformation process, strains of E. coli can incorporate extracellular plasmids into cytoplasm 'naturally' at low frequencies. A standard method was developed in which stationary phase cells were concentrated, mixed with plasmids, and then plated on agar plates with nutrients which allowed cells to grow. Transformed cells could then be selecte...
